Production of HPV16 capsid proteins in suspension cultures of human epithelial cells

Human papillomavirus (HPV) infection is a leading cause of morbidity and mortality in women worldwide. The virus associated with benign warts broad spectrum malignancies, including cervical cancer, considered disease high clinical relevance, especially developing countries. In this study we developed the production recombinant proteins HPV16 L1 L2 human cells suspension (293-F), which were transiently co-transfected pUF3L1h pUF3L2h vectors. Expressions capsid was detected by laser scanning confocal microscopy flow cytometry. Both identified intracellularly nucleus cytoplasm cells. presence these heterologous VLPs formation transmission electron (TEM) through colloidal gold immunolabeling negative staining. Cell extracts containing purified affinity chromatography immunization Balb/c mice formulation L1/L2 adjuvant able to induce higher titer anti-HPV16 L1, when compared antibodies indirect ELISA assay. These data indicate that transient expression 293-F efficiently established. results are promising for obtain HPV future studies involving papillomavirus, as well contribute development other vaccine strategies prevention against HPV.
